Mount BCIP/NBT-developed IHC slides with Immu-Mount (Thermo)?
I have been mounting my immunocytochemistry slides (that I have developed using BCIP/NBT AP) with Xylene-based PerMount, but color crystals form after a while and I can't use the data. I was wondering, as an alternative, could I mount these slides with Immu-Mount from Thermo as a permanent mounting media that will not cause crystals or any other bad side-affects to form? The Immu-Mount data sheet doesn't say anything about BCIP/NBT, yes or no, so I am not sure.
I would assume yes, you can use it...But all the microscopy I do is faded after a couple of days no matter what, so I never worry about long term storage. I get to the microscope, take hundreds of pictures and throw my slides out.
How long is a while? Do you hold onto your slides for a month, or does it happen during the drying process of the permount. The permount could be solubalizing something in your stain, which then crystalizes as the permount dries, so it might be good to move from an organic based mounting media to aqueous. Find a section that is not important and give it a shot.
